Measuring scale with 0.1 mm divisions The E10S 10x eyepiece is equipped with a scale in the plane of the field diaphragm. It serves as a normal eyepiece with 10x magnification and can also measure the size of objects or their elements. The scale is marked in millimeters and microns. To take measurements, the eyepiece must first be calibrated for each objective that will be used. The E10S eyepiece fits into a 23.2 mm tube.
Calibration
To perform calibration, you will need a calibration slide with a scale division of 0.01 mm. Place the slide on the stage and focus the microscope on its scale. Then, rotate the eyepiece in the tube until the divisions of the scales on the slide and the eyepiece are parallel to each other. Next, a calculation must be performed.
Calculation
You need to calculate how the divisions of the eyepiece and calibration slide (a stage micrometer) scales relate to each other: how many eyepiece scale divisions fit into the slide scale (for low magnification objectives) or how many slide divisions fit into the eyepiece scale (for medium or high magnification objectives). Then, use the following formula: E=TL/A, where: E = eyepiece scale division; T = stage micrometer scale division marked on the stage micrometer (0.01 mm); L = number of divisions on the stage micrometer; A = number of divisions on the eyepiece scale.
Record the number obtained as a result of the calculations for each objective.
The size of the samples is calculated as follows: first, determine how many divisions of the eyepiece scale it occupied. Then, multiply this number by the result obtained from the formula for each objective. This number will reflect the linear value of the sample.
