<p><strong>MAGUS CS5 Calibration Slide (XY0.01/0.07/0.15/0.6/1.5)</strong><br></p><p>X/Y micrometer scales with a 0.01mm scale division and circles 0.07/0.15/0.6/1.5mm in diameter </p></p><p>A calibration slide for microscopes.<br />The slide has four etched circles with diameters of 0.07mm, 0.15mm, 0.6mm, and 1.5mm. In the center of the slide, there are reticles: horizontal and vertical micrometer scales of 100 divisions. The scale division is 0.01mm, and the length of each scale is 1mm.<br />The calibration slide (stage micrometer) is used to determine the magnification, the microscope’s linear field of view, and the division value of the eyepiece scales and reticles.<br />With a digital camera, the slide is used to calibrate image analysis software for each objective.</p><br><p><strong>Image analysis software calibration</strong></p><br><p>Calibration is performed for each objective that will be used in observations. The calibration slide is used to determine the calibration factor for the image analysis software.</p><br><p>Calibration procedure:</p><br><ol><li>Fasten the calibration slide to the microscope stage.</li><li>Select the desired objective and adjust the maximum camera resolution.</li><li>Get a contrast image of the stage on the monitor screen and capture the image.</li><li>Select the “Calibration” mode in the image analysis software.</li><li>Click the mouse to define the maximum visible distance between the first and last scale division.</li><li>Enter the resulting scale size in real units into the image analysis software.</li><li>Enter the calibration name specifying the current objective magnification.</li><li>Check the result: in “Measurement” mode, measure a few divisions of the calibration slide scales in real units.</li><li>The image analysis software will save the calibration factor under the selected name. After that, it will automatically convert the results to the selected measurement units.</li></ol><br><p><strong>Eyepiece scale calibration</strong></p><br><p>The eyepiece scale calibration is performed for each objective that will be used in observations. The calibration slide should be placed on the stage, and the microscope should be focused on the measuring scale. The eyepiece should be rotated in the eyepiece tube until the scales on the slide and the eyepiece are aligned parallel.</p><br><p>If using low-power objectives, you need to calculate how many divisions of the eyepiece built-in scale fit into the slide’s measuring scale.<br />If using medium- and high-power objectives, you need to calculate how many divisions of the slide’s scale fit into the eyepiece scale.</p><br><p>The calculation for each objective is performed according to the formula:<br />Е=ТL/A, where<br />E – eyepiece division value<br />Т – stage division value specified on the stage micrometer (0.01mm)<br />L – number of stage micrometer divisions<br />А – number of eyepiece scale divisions.</p><br><p>The resulting values of calculating the eyepiece scale division for each objective should be written down.</p><br><p>When measuring an object, you should first count the number of eyepiece scale divisions the object spans. The result is multiplied by the previously calculated scale value for the selected objective. The resulting value will be the true linear value of the object.</p>
